A randomized, double-blind study by Hoffman et al.5 used ten healthy participants (5 males and 5 females) to measure the acute thermogenic effect of a weight loss supplement. Briefly, participants were required to report the laboratory on two separate occasions. Each time they were randomly asked to ingest 3 capsules of weight loss supplement or placebo. Thereafter, resting oxygen uptake, respiratory quotient, caloric expenditure, heart rate and blood pressure measures were taken at regular intervals until 3 hours post ingestion. In addition, the effects on the mood were assessed by the Profile of Mood States (POMS) questionnaire. Statistical analysis revealed significant differences in oxygen uptake and energy expenditure between the experimental and placebo groups. The study also reported a trend towards a greater use of stored fat as a fuel. Psychological data showed no significant difference between the groups except for tension and confusion variables. Overall, this study concluded that there was a significant increase in energy expenditure in young, healthy individuals following acute use of a weight loss supplement containing the aforementioned ingredients.5 Given the effectiveness of these ingredients, such results are expected. For example, Pausinystalia Extract is also one of the ingredients used in the Slimexy™ formula. McCarty7 suggested that Pausinystalia johimbe extract taken prior to exercise can increase lipolysis and serum free fatty acids (FFA) levels during and following exercise. This promotes fat loss and therefore the efficacy of exercise training is enhanced.7
Bloomer et al.2 tested further effects of the same weight loss supplement as in Hoffman et al.’s study5. A sample size of 20 healthy participants (10 men, 10 women) was used in this randomized, placebo controlled, cross-over study. Compared to previous similar studies, Bloomer et al. measured a more prolonged as opposed to just acute effects by taking samples up to 6 hours post ingestion. Biochemical and metabolic measures included plasma norepinephrine, epinephrine, glycerol, FFA and metabolic rate. Similar to Hoffman et al.’s5 findings, results demonstrated an increase in catecholamine secretion, lipolysis, and metabolic rate. The effectiveness was reported to be similar in males and females.
The same research group investigated the effects of the weight loss supplement in just males.3 Ten healthy, resistance trained men participated in the randomized, double-blind, cross-over design study. Markers of lipolysis, metabolic rate, hemodynamics and blood catecholamine levels were measured for 90 minutes post ingestion of either 3 capsules of the weight loss supplement or placebo. There was an acute increase in plasma norepinephrine, glycerol, FFA and metabolic rate in the experimental group. Results also showed a slight increase in heart rate and systolic but not diastolic blood pressure.3 Increases in plasma norepinephrine have been linked with the ability to stimulate fatty acid mobilization. Interestingly, the blood variables peaked 90 minutes after ingestion of the weight loss supplement. Therefore, it is likely that positive effects on various markers last longer than just 90 minutes. In addition, the positive results on metabolic rate and lipolysis were observed in the sample of young, healthy and lean men. Thus, it can be speculated that even greater positive changes would be observed in overweight/obese sedentary population.3
Similar to Bloomer et al.3, Jitomir et al.6 conducted a double-blind, randomized, placebo controlled, cross-over design study using twelve healthy male participants. Compared to previous studies analyzing the effects of the weight loss product at rest, this study aimed to determine whether similar changes are observed when supplementation is combined with exercise. Participants were tested before, during and after exercise. The testing protocol included taking regular measurements of resting energy expenditure (REE), respiratory exchange ratio (RER) to assess fuel oxidation, heart rate and blood pressure. Participants were asked to perform a maximal oxygen uptake test on a treadmill in order to monitor responses not only pre- and post-exercise but also during it. One of the main findings was that REE values were significantly elevated up to 120 minutes post-ingestion. RER values remained decreased for up to 60 minutes post-ingestion demonstrating a greater fat oxidation. Interestingly, no significant differences in these measures were observed between the experimental and placebo groups after the maximal exercise. Finally, the supplement did not have a significant effect on heart rate and blood pressure values before, during or after exercise.6
